Registration of 252 sequenced sorghum mutants as a community reverse genetic resource

چکیده

Induced mutagenesis is a powerful approach to generate variations that can be used toward the elucidation of gene function and create new traits for crop improvement. We have developed pedigreed mutant library through chemical with ethyl methanesulfonate (EMS)-treated seed from sorghum (Sorghum bicolor L. Moench) inbred line BTx623. The displayed wide diversity phenotypes, including potential significant agronomic value. A selection 252 lines (Reg. no. GS-794–Reg GS-1045, PI 701562–PI 701813) were sequenced an average depth 16x paired-end sequencing using Illumina Highseq. This endeavor resulted in detection cataloguing more than 1.8 million canonical EMS-induced mutations (variants). variants distributed across genome uniformly, 24,757 genes affected by impactful may alter function. Mutations interest searched online SorghumBase (https://www.sorghumbase.org). These mutants ordered GRIN (https://www.ars-grin.gov/) no associated fee.

Retention of induced mutations in a Drosophila reverse-genetic resource.

Targeting induced local lesions in genomes (TILLING) is a reverse-genetic method for identifying point mutations in chemically mutagenized populations. For functional genomics, it is ideal to have a stable collection of heavily mutagenized lines that can be screened over an extended period of time. However, long-term storage is impractical for Drosophila, so mutant strains must be maintained by...

A Drosophila Genetic Resource of Mutants to Study Mechanisms Underlying Human Genetic Diseases

Invertebrate model systems are powerful tools for studying human disease owing to their genetic tractability and ease of screening. We conducted a mosaic genetic screen of lethal mutations on the Drosophila X chromosome to identify genes required for the development, function, and maintenance of the nervous system. We identified 165 genes, most of whose function has not been studied in vivo. In...

Genetic engineering of grain sorghum (Sorghum bicolor (L

PhD thesis Sorghum (Sorghum bicolor L. Moench), the fifth most important cereal crop in the world, is one of the most important staple crops in Africa and South Asia and represents the only viable food grain for many of the world’s most food insecure people. However, sorghum grain shares the typical nutritional deficiencies of cereal grains, a low content of several essential amino acids. There...
